Global First Non-Opioid Painkiller Approved, Marking a Milestone Breakthrough in Pain Management
On January 30th local time, the U.S. FDA announced the approval of a new drug, Suzetrigine, from Vertex for the treatment of moderate to severe acute pain in adults.
On January 30, 2025, the U.S. FDA officially approved Vertex's new drug, Suzetrigine (trade name JOURNAVX), for the treatment of moderate to severe acute pain in adults. This world's first novel non-opioid analgesic offers a new solution to address the pain of opioid addiction. Suzetrigine selectively inhibits NaV1.8 channels on peripheral pain-sensing neurons, blocking pain signal transmission and preventing the euphoria and addiction-inducing effects in the brain. Its efficacy has been validated in two randomized, double-blind trials. In patients undergoing abdominoplasty and bunionectomy, high-dose treatment improved 48-hour pain intensity scores (SPID48) by 48.4 and 29.3, respectively, compared to placebo, demonstrating good tolerability.
This drug's approval not only reshapes the acute pain management paradigm but also drives industry research and development focus. Currently, the wholesale price of 50mg tablets of Suzetrigine in the US is approximately 111.39 yuan per tablet. Domestic companies have accelerated their efforts in the same target market: Hengrui Pharma and Jemincare's NaV1.8 inhibitors have entered Phase II clinical trials, while products from companies like Joincare and Haibowei Pharma are in Phase I. Of the 30 active new NaV1.8-targeted drugs globally, 12 have advanced to clinical trials for pain indications, making non-opioid analgesia a hot topic in the industry.
From an industry perspective, the approval of Suzetrigine marks a shift in pain treatment from a "balance between analgesia and addiction" to "non-addiction analgesia." Globally, over 40 million Americans rely on opioids for acute pain each year, and 85,000 develop substance use disorders as a result. Non-opioid medications are crucial to addressing the opioid crisis. With new mechanism drugs such as dual receptor agonists (such as Tris Pharma's Cebranopadol) entering Phase III, coupled with the intensive layout of domestic and foreign companies in ion channel targets, the pain treatment track will accelerate the technological upgrade and market expansion.
